生於西班牙首都馬德里，幼時家中開辦一家理髮店；曾於西班牙國家音樂學院學習芭蕾舞專業。她的妹妹Mónica Cruz也是女演員，她的弟弟Eduardo Cruz為西班牙炙手可熱的歌手。 2010年7月，她與西班牙著名男演員巴爾登(Javier Bardem)結婚。2011年1月，產下一子Leonardo。 2007年，潘妮洛普·克魯茲憑電影《玩美女人》Volver奪得坎城電影節以及歐洲電影獎最佳女主角獎，並憑此片入圍奧斯卡、金球獎及英國電影學院獎最佳女主角。2009年，她憑電影《情遇巴塞隆納》Vicky Cristina Barcelona奪得奧斯卡金像獎以及英國電影學院獎最佳女配角獎，並憑此片入圍金球獎最佳女配角。2011年4月，入主好萊塢星光大道。
